چکیده

Dry powder formulations are often composed of fi ne drug particles and coarser carrier particles, typically -lactose monohydrate because it is an inert, cheap, broadly available and non-toxic excipient1. However, the effi ciency of a powder formulation may be highly dependent on the lactose quality and source2. Indeed, interactions between particles are mainly dependent on the physicochemical characteristics of the interacting particles. The aim of this work was to identify the characterisitcs of lactose that infl uence the drug-to-carrier interaction and the performance of dry powder formulation. For this purpose, we compared marketed lactoses obtained by milling or by sieving. From the same raw materials, 2 fractions of lactose without fi nes were also prepared and tested.

Examining the influence of carrier lactose on the performance of dry powder inhaler (DPI) formulations: A case study

Introduction Most commercially-available dry powder inhaler (DPI) formulations utilize a carrier-based formulation concept. Utilizing a mixing process, micronized active pharmaceutical ingredient (API) particles are attached to the surface of larger excipient particles. Multiple studies list excipients such as mannitol, glucose monohydrate, trehalose, dextrose, maltose, sorbitol and maltitol fo...

Development of an Inhaled Sustained Release Dry Powder Formulation of Salbutamol Sulphate, an Antiasthmatic Drug

The present research was aimed to develop and characterize a sustained release dry powder inhalable formulation of salbutamol sulphate. The salbutamol sulphate microparticles were prepared by solvent evaporation method using biodegradable polymer poly (D,L-lactic-co-glycolic acid) to produce salbutamol sulphate microparticle mixed with carrier respirable grade lactose for oral inhalation of dry...

The following manuscript entitled “An investigation into the effect of fine lactose particles on the fluidization behaviour and aerosolization performance of carrier-based dry powder inhaler formulations” has been published in AAPS PharmSciTech 2014 Aug;15(4):898-909

The effect of milled and micronized lactose fines on the fluidization and in vitro aerosolization properties of dry powder inhaler (DPI) formulations was investigated, and the suitability of static and dynamic methods for characterising general powder flow properties of these blends was assessed. Lactose carrier pre-blends were prepared by adding different lactose fines (Lactohale 300, 230 and ...

Lactose engineering for better performance in dry powder inhalers.

Dry powder inhaler (DPI) is generally formulated as a powder mixture of coarse carrier particles and micronized drug with aerodynamic diameters of 1-5 μm. Carrier particles are used to improve drug particle flowability, thus improving dosing accuracy, minimizing the dose variability compared with drug alone and making them easier to handle during manufacturing operations. Lactose is the most co...

Prediction of dry powder inhaler formulation performance from surface energetics and blending dynamics.

The purpose of these studies was to investigate the ability of surface energy measurements and rates of mixing in dry powder inhaler (DPI) formulations to predict aerosol dispersion performance. Two lactose carrier systems comprising either spray-dried or milled particles were developed such that they had identical physical characteristics except for surface morphology and surface energies avoi...
